ABSTRACT

The aim of this research was the development of [111]In-labeled porphyrins as possible radiopharmaceuticals for the imaging of tumors. Ligands, 5, 10, 15, 20-tetrakis [3, 5-dihydroxyphenyl] porphyrin] [TDHPP], 5, 10, 15, 20-tetrakis [4-hydroxyphenyl] porphyrin [THPP] and 5, 10, 15, 20-tetrakis [3,4-dimethoxyphenyl] porphyrin] [TDMPP] were labeled with [111]InCl3 [produced from proton bombardment of natCd target] in 60 min at 80[degree sign]C. Quality control of labeled compounds was performed via RTLC and HPLC followed by stability studies in final formulation and presence of human serum at 37[degree sign]C for 48 h as well as partition coefficient determination. The biodistribution studies performed using tissue dissection and SPECT imaging up to 24h. The complexes were prepared with >99% radiochemical purity [HPLC and RTLC], high stability in 48h. Partition coefficients [calculated as log P] for [111]In-TDHPP, [111]In-THPP and [111]In-TDMPP were 0.88, 0.8 and 1.63 respectively. Due to urinary excretion with fast clearance for [111]In-TDMPP, this complex is probably a suitable candidate for considering as a possible tumor imaging agent

ABSTRACT

An eleven year old boy presented with headaches and dizziness associated to micturition. On radiologic imaging, he was found with a bladder mass. The biochemical work up was suggestive of pheochromocytoma. An OctreoScan (111In-pentreotide) was used to rule out metastatic extension or other extra-adrenal locations of the pheochromocytoma. OctreoScan data correlated well with other radiologic studies, operative findings and with the final diagnosis, validating its use on pediatric patients.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of 90Y-DOTATOC and 131I-MIBG in treatment of metastatic medullary thyroid carcinoma (MTC).</p><p><b>METHODS</b>Twelve histologically confirmed patients with metastatic MTC were included. All patients underwent both 111In-DTPA-octreotide imaging and 131I/ 123I-meta-iodobenzylguanidine (MIBG) imaging. According to the results of the combined imaging, positive patients were selected to be treated with 90Y-DOTA-D-Phe1-Tyr3-octreotide (90Y-DOTATOC) or 131I-MIBG, respectively. The therapeutic procedures of targeted internal radiation were performed with 3.33 GBq 90Y-DOTATOC at 6-week intervals, or 11.1 GBq 131I-MIBG with a minimum interval of three months.</p><p><b>RESULTS</b>The imaging procedure was positive in all 12 patients: 111In-DTPA-octreotide imaging in eight patients, 131I/ 123I-MIBG imaging in six patients. According to the results of combined imaging, we identified four patients to be treated with 90Y-DOTATOC, and five patients with 131 I-MIBG. After three to five sessions of treatment, three patients with partial remission and six with stable disease were observed. The effective rate was 3/9 (33.3%) and the overall tumor response rate was 9/9 (100%). No relevant toxicity was observed.</p><p><b>CONCLUSION</b>The combined imaging technique can be used to identify patients for effective radionuclide treatment. The treatment with 90Y-DOTATOC or 131I-MIBG is well tolerated and may improve the fate of patients with metastatic MTC.</p>

ABSTRACT

The beta 2-adrenoceptor agonist salmeterol inhibits the accumulation of eosinophils at the site of allergic inflammation, but the cellular target is uncertain. This study was undertaken to determine whether eosinophils themselves are the target of this inhibitory action. Purified guinea pig peritoneal eosinophils were labeled with indium-111 oxine, pre-incubated with salmeterol or drug vehicle before being injected intravenously into guinea pigs, which have previously been immunized with ovalbumin [OA]. Four hours after intradermal injection of OA or platelet-activating factor [PAF], the accumulation of labeled eosinophils at the injection sites was determined by measuring the radioactivity in punched-out skin sites. In vitro adhesion to plastic plate was also studied by measuring the eosinophil peroxidase content of adhering cells. About 6% of the injected 111In-eosinophils were circulating 10 min after injection and remained relatively steady for over 4 h. In animals given vehicle-treated 111In-eosinophils an up to 3.6-fold increase in the accumulation of the labeled cells at the skin sites of OA or PAF injection was seen. Pretreatment of 111In-eosinophils with salmeterol [1 micro M] - a concentration giving about 65% inhibition of in vitro adherence - had no effect on the basal [PBS-induced] skin accumulation of the injected cells. However, it inhibited the net accumulation induced by OA [0.01-1 micro g/site] and PAF [0.01-1 nmol/site] by 58.8-100%. At 1 micro M, salmeterol itself had no significant effect on the viability and circulation of 111In-eosinophils. These results provide evidence for a direct inhibitory effect of salmeterol on eosinophils and suggest that this may account for a significant part of its clinical anti-inflammatory properties

ABSTRACT

El objetivo de este estudio ha sido comparar la capacidad de la gammagrafia con citrato de Gálio-67 (67Ga), de los leucocitos marcados con Tc-99m (99mTc-leucocitos) y de la inmunoglobulina G marcada con In-111 (111In-IgG) en el diagnóstico de infecciones óseas y articulares. Han sido estudiados 56 pacientes. En todos ellos se indicó una gammagrafia ósea (99mTc-MDP). Los pacientes han sido aleatoriamente asignados para estudios con 67Ga (Grupo 1 - 22 pacientes) o con la administración simultánea de 99mTc-leucocitos y con 111In-IgG.(Grupo 2 - 34 pacientes). Para los grupos 1 y 2 han sido confirmadas 12/26 y 11/34 lesiones sépticas respectivamente. La gammagrafia con 67Ga mostró 6 verdadero-positivos, 9 verdadero-negativos, 5 falso-positivos y 6 falso-negativos. Con los 99mTc-leucocitos, los resultados han sido 8, 18, 5 y 3, respectivamente. Con la 111In-IgG, 7, 17, 6 y 4, respectivamente. La sensibilidad, especificidad y precisión han sido respectivamente del 50 por ciento, 64,3 por ciento y 57,7 por ciento para el 67Ga, 72,7 por ciento, 78,2 por ciento y 76,4 por ciento para los 99mTc-leucocitos y 63,6 por ciento, 73,9 por ciento y 70,6 por ciento para la 111In-IgG. En este estudio, la gammagrafia con los leucocitos marcados presentó mayor precisión que con 67Ga y con 111In-IgG en el diagnóstico de infecciones óseas y articulares. Sin embargo, en el diagnóstico de infecciones en prótesis articulares los resultados han sido menos fiables

ABSTRACT

Se revisa la utilidad de la ventriculografia nuclear en el estudio de la funcion y de los volumenes telediastolicos y telesistolicos de ventriculo izquierdo en los pacientes con miocardiopatias dilatadas, hipertroficas y restrictivas. Tambien se valora el aporte de los estudios al seguimiento de pacientes a quienes se les realiza una miocardioplastia dinamica, incluyendo la determinacion del indice de regurgitacion o relacion entre las amplitudes ventriculares. En el caso de la miocarditis, enfermedad especifica del musculo cardiaco, se comentan los metodos de deteccion de dano miocardico y se hace hincapie en el empleo de la gammagrafia con anticuerpos monoclonales antimiosina
